ELISpot and FluoroSpot
ELISpot and FluoroSpot are highly sensitive functional assays for measuring antigen-specific T cells, which typically occur at low frequency in vivo. Crux Biolabs offers both ELISpot and FluoroSpot assays to support R&D and pre-clinical to clinical projects providing validation services using commercially available or custom assays.
ELISpot
ELISpot assays provide robust measurement of measuring the frequency of cytokine-secreting cells at the single-cell level. Here target cells are cultured on a surface coated with a specific cytokine capture antibody in the presence or absence of stimuli. Cytokines are visualised using chemiluminescence, with each spot corresponding to a single secreting cell.

FluoroSpot
FluoroSpot assays are like ELISpot assays but rely on detection using fluorescently labelled detection antibodies. Fluorescent detection enables simultaneous measurement of different analytes secreted at a single-cell level. This allows parallel detection of up to four cytokines secreted by immune cells upon antigen recognition.
